The Inductions of Tyrosine Aminotransferase by Glucagon and Hydrocortisone*
نویسندگان
چکیده
Glucagon alone, given to adrenalectomized rats, caused induction of tyrosine aminotransferase only in repeated doses. A single dose given with or 5 hours after hydrocortisone enhanced 3-fold the induction of tyrosine aminotransferase by the hydrocortisone. Glucagon did not alter the levels of tryptophan oxygenase in these situations. The simultaneous administration of actinomycin D completely inhibited the effects of glucagon on tyrosine aminotransferase when glucagon was given alone, with hydrocortisone, or after hydrocortisone. The induction of tyrosine aminotransferase by glucagon both in untreated rats and in rats treated by injection of hydrocortisone therefore appears to require synthesis of ribonucleic acid; this requirement is distinct from the RNA synthesis required in the induction of the same enzyme by hydrocortisone. Actinomycin given with the hydrocortisone prevented its induction of tyrosine aminotransferase and tryptophan oxygenase, but actinomycin given more than 1 hour after hydrocortisone did not decrease or increase the levels of these enzymes.

Regulation of tyrosine alpha-ketoglutarate transaminase in rat liver. 8. Inductions by hydrocortisone and insulin in cultured hepatoma cells.
Hydrocortisone addition to cell cultures of the Reuber (H-35) hepatoma initiated an 8to IO-fold increase in tyrosine transaminase (L-tyrosine:Z-oxoglutarate aminotransferase, EC 2.6.1.5) levels, the response being maximal at 9 hours and persisting at this new level for at least 36 hours. The optimal concentration of hydrocortisone for this induction was 5 X low7 M. Insulin was also found to ele...

متن کاملInitiation by glucagon of the premature development of tyrosine aminotransferase, serine dehydratase, and glucose-6-phosphatase in fetal rat liver.
The explosive accumulation of tyrosine aminotransferase that normally occurs in rats at birth has been brought about prematurely by a single administration of glucagon to fetuses. Serine dehydratase is also found to accumulate normally after birth, but glucagon initiates its formation prenatally. The glucose-6-phosphatase activity of liver, which begins to develop just before birth, has been pr...

In primary cultures of rat hepatocytes, addition of dexamethasone (10 microM) plus glucagon (0.5 microM) caused several-fold increases in the activities of serine dehydratase (EC 4.2.1.13), tryptophan oxygenase (EC 1.13.11.11), and tyrosine aminotransferase (EC 2.6.1.5) in 24 h. These inductions were inhibited by insulin. Addition of epinephrine or phenylephrine at 10 microM blocked these induc...
متن کاملHormonal regulation and the effects of glucose on tyrosine aminotransferase activity in adult rat hepatocytes cultured on floating collagen membranes.
Adult rat parenchymal hepatocytes can be maintained in primary culture on floating collagen membranes of prolonged periods of time. In this system the enzyme tyrosine aminotransferase is induced by glucagon, (10(-6) to 10(-8) M) hydrocortisone (10(-5) to 10(-8) M), and cyclic adenosine 3':5'-monophosphate (cAMP) (10(-4) to 10(-5) M). Epinephrine (10(-4) M) induces the enzyme only in the presenc...
